Crystallographic Grade Recombinant Human RENIN bulk quantities from 10-500mg single lot sizes. customs fills for human recombinant renin available.
Crystallographic Grade Recombinant human pro-renin was expressed in HEK cells, converted to renin, an active enzyme was purified using peptide affinity chromatography.
Human recombinant Renin Activity was measured by the cleavage of a fluorescent peptide substrate (Arg-Glu(EDANS)-Ile-His-Pro-Phe-His-Leu-Val-Ile-His-Thr-Lys(dabcyl)-Arg) from Molecular Probes (Cat # R-2931) and Renin activity was detected in a microplate format using a SpectroMax Gemini microplate reader. recombinant human Renin activity was measured as ~100 FU/min/g protein at the Km. Recombinant human Renin protein concentration was determined using amino acid analysis

What is Human Renin?
Human Renin, also known as angiotensinogenase, is a circulating enzyme released mainly by juxtaglomerular cells in the JGA of the kidneys in response to low blood volume or low body NaCl content, mediated through the release of prostaglandins. Although human Renin has hormone-like actions, human Renin cleaves a protein precursor in the circulation rather than working on a cellular target.
Human Renin is an important enzyme in the regulation of blood pressure and electrolyte homeostasis and therefore is of great clinical importance. Kinetic and mechanistic functions of human Renin are of importance for the development of hypertension drugs. Plasma renin activity (PRA) is measured as part of the diagnosis and treatment of hypertension.
Human Renin acts as a proteolytic enzyme of the kidney by releasing angiotensin which has a vasoconstrictive action which results in a higher blood pressure. Early detection of Human renin level increases related to a blood pressure increase have shown in research to provide vital prognostic and diagnostic guidelines for evaluating possible surgical treatment especially in renovascular involvement.

Human Renin Study: Human Renin is synthesized as an enzymatically inactive prorenin, which is split by limited proteolysis to give the active human renin enzyme. Both human renin and human prorenin is secreted by the kidneys. Circulating human renin concentrations are occasionally raised in nephroblastoma.
